these is assignment rule I have created 

but its not populating the assign to on incident form :

Is these users part / member of these groups?

Then it will noy work, bcz assigned to is depend on assignment group.

and also it is illogical that you are assigning to different user and group is different.

but here is member is not part of that assignment group
