I want to populate the location field by discovery to update it in CMDB.

Discovery is unable to determine the location.
It's possible to add a location on the Discovery Schedule and this is used to populate the location field on the CI. But this means that you need a schedule for each location.
Another solution could to create a custom table were you map subnets to locations. When a device with IP address is discovered a Business Rule can be used to check the custom table to find the location based on the IP address.

You can define location on the Discovery Schedule. If you split your schedules by IP range and location, the location will populate to the CIs found in each one.
