Example default Database group, manager is Don Goodliffe.
Change -> Create New.
Set Assignment group to Database.
How to change approval from Assignment group to Manager of Assignment group?

Ahhhh I see what you are trying to do.
My bad I should have corrected you for that. Approval- Group will fire off the approval from all the members so if you want approval from either of the members , then use this functionality. Not sure why you don't have a approval from Don Goodlife. Try adding that to the members related list of the group and then see. Strange that it just sees the members as opposed to should see manager too.
If you want specific members , then use User Approval instead and do the dot walking over there.

That is because the manager is not a member of the group.
Group approval looks for the members.

"The Approval - Group activity creates approval records for each member of a specified group."
It is possible to be the manager without being a group member
And you cant selected assignment_group.manager because you are using group approval and not user approval

Change it to user approval and choose assignment_group.manager
