Form layout is simple and old way of adding fields on form.

Form Layout is the simple style editor where you can add, remove, or reorder fields of form

Form Design and Form Builder is drag-and-drop where you can visually arrange fields, create sections, and build multi-column layouts. 

As from prompt you can see ServiceNow is recommending Form Builder now a days

These are like different interfaces to work on a form. Form Builder is new, and it gives more drag-and-drop options and easier creation of new fields. You can also see the design and how a field will look on the form. So, in short, Form Builder is like a combination of Form Designer and Form Layout.
