Hi Friends,
I would like to understand more on company separation. can any one please provide more details and confirm whether company separation can be configured in single instance (without domain separation)?

To be clear Domain separation is a single instance. MSPs are issued a single (production) instance and use Domain Separation to keep their client data separated while being able to leverage the same logic (or provide custom logic for different client processes.) This typically isn't needed outside the MSP space and we don't recommend it for single customer instances as it causes a lot of overhead. Please work with your ServiceNow account team to determine if it is right for you. As stated before, once it's turned on, there's no going back and if you are not an MSP, then it's probably not the right solution for you. There are rare exceptions, but work with the team to make sure.
