We recently udated our now assist for ITSM plugin to the newest version.
After the update we no longer have access to Now Assist Skill Kit.
Before the update we can access it just fine.
I do have the sn_skill_builder.admin role.
Some of our application runs on now assist skills we made ourselves but now all of them are not working.

Indeed, we had to do further steps to get your issue resolved based on another defect documented in KB2688829. If the workaround in this KB doesn't fix the issue, one needs to raise a support case to implement another workaround that can only be done by Support, which is exactly what you did. The issue should now be resolved, as your case raised yesterday is in SP.

This seems to be a role issue. Can you check the if any ACLs were added and are causing the problem? Else log a Now support case as it is issue after update.

Please review KB2734243: After upgrading "AI Agent Studio", "Now Assist Skill kit" & "Now Assist Admin" not visible with security constrains error. I found this article using the Now Assist (Virtual Agent/Support Portal Search) on the ServiceNow Support Portal.
