Once you have a license giving access to App Engine Studio (for instance, App Engine Enterprise) you are entitled to use it.
Using it however, does not consume an App Engine license as such - you consume those licenses by:
* Creating tables and assigning those tables to App Engine Enterprise license
* Creating roles and giving those roles access to the above table(s) on a Fulfiller level.
* Assigning above role(s) to a User (likely by adding Role to Group where User is Member).
