If Manager was left from Organization, How we can design workflow?

Hi, unfortunately your question is not clear.
If the requirement is to design dynamic workflow IE not to hard code the manager value in the workflow.
Then you can reference any 'user' reference field on a record,or dot.walk to any related user field eg group.manager
