How can we set a tool-tip for topics in Megamenu. Is there any OOB functionality/any custom steps which we can follow. Any Insights would be helpful.

Unfortunately out-of-the-box no, no configuration available for this.

You might achieve this customizing the widget, though that will cost some time and energy, and you need to maintain the widget as such widgets are updated often by ServiceNow and else you will miss important or useful updates.
