I need to know how count license used in HR Service Delivery application. Assigned Roles, entry in HR Profile or other?

HR is based on a user subscription model, not fulfiller/role based. License usage is based on the count of active users in the HR profile table.

And then click the Role you're interested in knowing the count for, for example ITIL, select it and then look at the users tab:

Our licensing model has changed over the years so I want to provide further clarification for anyone that may reference back to this question. Current licensing is based on employee count using active HR Profiles, as defined by the user they are associated to being active and/or today's date being within their start and end dates.
Legacy licensing was on a fulfiller model where the customer purchased additional licenses for their agents/fulfillers. This licensing was associated to only case management and is not licensed for any of the newer application such as Employee Service Center, Enterprise Onboarding & Transitions, and Employee Document Management.

HR License include approver role. If I assign to a users in HR Profile also a approver role need to pay?
