I have created a order guide for employee onboarding in which I am asking employee to add manager in the manager fields. request should go the selected manager for approval and on approval all the requests should get approved automatically.

I meant add manager in the order guide and then that manager approves request

So, you want to select the manager using an Order Guide variable, and then pass that manager to the Request flow for approval — is that correct?
You can cascade the Order Guide variable to the REQ/ITEM level, and then use it within the flow.

that order guide contains which all catalog items?
which other REQs should get auto approved?
