If a user click unsubscribe in the email notifications sent by service now
will he get the mails or nor ....?
If not how he will subscribe again...?

The use can update his notification preferences to receive the mails again. To see the notification preferences, he can go to Profile -> Preferences -> Notifications , then enable Corresponding Notification type.

Unsubscribe means, user will not get that email in his./ her mail box and yes he/she can subscribe again.
