Lob is a custom field but for customer and business service fields the details are available in service mapping so when we run a particular discovery pattern it should populate, but it isn't?

Those fields are not discoverable on the servers. They are not coded into any setting.

If all of your Servers have the same LOB then you can set that as a default value. Otherwise you will have to perform a lookup somewhere to supplement those values.

What about Customer & Business Service fields? Can we auto-populate that?

I am not sure about what your Customer field means to your organization, so I will skip that. 

For Business Service, you can map a CI to a Service using a dynamic CI grouping or actually implementing Service Mapping. My guess is that you simply just want to provide the value not actually map things out.

If your servers have naming convention that can easily identify the service, then you can automate that as well.

How do you know which server goes with which business service?

If you can answer this - Technically it can be automated . I doubt these fields are discoverable , these seems more of a ORG Specific Logical CI.
